ACE-I block the conversion of angiotensin 1 to angiotensin II, which causes vasoconstriction and stimulates the production of aldosterone synthesis. Thus, ACE-1 promote vasodilatation and decrease sodium retention, consequently lowering blood pressure. The inhibition of angiotensin-converting enzymes also blocks the breakdown of bradykinins. The increase in bradykinin level leads to additional vasodilatation effects. However, the increase in bradykinin is also responsible for adverse effects such as cough and angioedema. [Pg.25]

Angiotensin antagonists (Angiotensin-converting enzyme inhibitors) and ACE inhibitors lower blood pressure and are used to treat heart failure by causing the excretion of sodium and water. ACE inhibitors block the release of aldosterone. Aldosterone causes the retention of sodium. Cardiac output and heart rate remain unchanged. ACE inhibitors are administered to patients with elevated serum rennin levels. ACE inhibitors can cause angioedema in which the mouth, throat, lips, eyelids, hands, and feet swell. [Pg.299]

Angiotensin-converting enzyme inhibitors are effective in the treatment of hypertension and congestive heart failure. ACE inhibitors also prevent bra-dykinin metabolism and, as a result, may produce the side effect of cough and angioedema. Many of the known ACE inhibitors were developed without the aid of computational chemistry techniques.
