Amine synthesis from reductive amination of a ketone and an amine in the presence of excess formic acid, which serves as the reducing reagent by delivering a hydride. When the ketone is replaced by formaldehyde, it becomes Eschweiler-Clarke reductive alkylation of amines. 

This elegant synthesis uses some of the methods of amine synthesis from this chapter and looks forward to the next chapter on protecting groups as well as later discussion of nitro group chemistry.

Secondary Amine Synthesis from Primary Amines. ... 

Nitroarenes and nitriles could also be employed as amine precursors. In 2010, Li and co-workers reported a ruthenium complex-catalyzed synthesis of tertiary amines by A -alkylation of nitroarenes with alcohols (Eq. 14) [87], In this method, large excess amounts of the alcohols (mostly 7.5 equiv.) are necessary to reduce the nitroarenes to anilines prior to A -alkylation. In 2011, Shi and co-workers also developed an amination reaction for secondary amine synthesis from nitro or nitrile compounds (Eq. 15) [88]. In the same year, Deng and co-workers reported a ruthenium-catalyzed method for tertiary-amine synthesis from nitriles and primary alcohols [89]. In 2013, Beller and co-workers reported another A -alkylation reaction of nitrile compounds with secondary alcohols [90].

Another important reaction of diketene derivatives is the Hant2sch pyridine synthesis (101). This synthesis is the preparation of 1,4-dihydropyridines (14) starting either from two acetoacetic esters, which react with an aldehyde and ammonia or a primary amine or from 3-aminocrotonates and 2-alkyhdene acetoacetic esters, both diketene derivatives. Several such dihydropyridines such as nifedipine [21829-25-4] (102), nimodipine [66085-59-4] and nicardipine [55985-32-5] exhibit interesting pharmaceutical activity as vasodilators (blood vessel dilation) and antihypertensives (see Cardiovascularagents). 

Synthesis and Manufacture of Amines. The chemical and busiaess segments of amines (qv) and quaternaries are so closely linked that it is difficult to consider these separately. The majority of commercially produced amines origiaate from three amine raw materials natural fats and oils, a-olefins, and fatty alcohols. Most large commercial manufacturers of quaternary ammonium compounds are fully back-iategrated to at least one of these three sources of amines. The amines are then used to produce a wide array of commercially available quaternary ammonium compounds. Some iadividual quaternary ammonium compounds can be produced by more than one synthetic route.
