The majority of hK3 (PSA) in the prostate and seminal plasma is in its free form, with less than 5% complexed with protein C inhibitor [111]. In serum, the majority of hK3 binds to protease inhibitors, and only 15%-25% is in the free form. The hK3-ai-antichymotrypsin complex constitutes 70%-85% of total serum hK3, with the hK3 2-macroglobulin and hK3 o,-antitrypsin complexes representing 15% and 3%, respectively [112-114],...

Prostatic Carcinoma. Plasma levels of AMG are depressed pretreatment in advanced carcinoma of the prostate and return to normal with successfiil treatment. AMG binds to prostate-specific antigen (PSA), and levels of the complexes are elevated in plasma during active disease, as is the case for PSA-ai-antichymotrypsin complexes. In vitro, PSA added to serum has a higher affinity for AMG than for tti-antichymotrypsin. In vivo, the complexes are probably removed very rapidly by the liver, as are other protease complexes with AMG usually, free PSA and PSA-tti-antichy-motrypsin are the only forms detected. 

Kallistatin, a human tissue kaUikrein ( kaUikreins) inhibitor and an additional member of the serpin superfamUy that inhibits human tissue kaUikrein. The translated sequence of kaUistatin (Mr 58 kDa) shares about 45% sequence identity with, for example, human ai-antichymotrypsin, protein C inhibitor, and ai-antitrypsin. The unique reactive site Pl-Pl of kaUistatin is Phe-Ser. KaUistatin occurs in the plasma at a average level of 13.5 mg The recombinant kaUistatin produced in E. coli is very simUar to the native inhibitor in its biochemical properties, despite the lacking glycosylation [K. X. Chai et al.,/. Biol. Chem. 1993, 268, 24498 J. L. Chao et al., /. Lab. Gin. Med. 1996, 127, 612].
