While the "microsomal" fraction of most oilseeds is highly active with respect to TG biosynthesis, the role of other subcellular fractions is less well documented. In this report the properties and subcellular distribution of the acyltransferases responsible for channelling acyl moieties either to PC or to the Kennedy pathway in oilseed rape are described. [Pg.537]

Microsomal fractions of oilseed rape readily incorporated (1- C) oleoyl CoA into PC, PA and TG, indicating the presence of active Kennedy pathway and PC-acyltransferase activities. Acyl transfer to PC was hugely stimulated in the presence of exogenous LPC but, in the presence of exogenous LPA the vast majority of the acyl transfer was to PA (Table 1). In each case the acylation of the added lysophospholipid was virtually the only route for oleate metabol-isation, resulting in the accumulation of labelled PC or PA over the short term. By means of pulse-chase studies, it was possible to observe some flow of label from PA towards TG and PC, which is consistent with the proposed operation of an equilibrium between DG and PC (1-4). Photoreactive acyl-CoA analogs were used in the characterization of diacylglycerol acyltransferase (DGAT, EC 2.3.1.20) from microspore-derived (MD) embryos of oilseed rape. 12-[(4-Azidosalicyl)amino]dodecanoyl-CoA (ASD-CoA) was more effective than 12-azidooleoyl-CoA in the inhibition of DGAT activity. Photoaffinity labeling of particulate and solubilized fractions with 25l-labeled ASD-CoA resulted in the incoiporation of label into a number of polypeptides but fewer polypeptides were labeled in the solubilized fraction. Thus, photoaffinity labeling may be useful in the purification and characterization of DGAT. [Pg.518]
