A variety of l,2,3,4-tetrahydro-j8-carbolines have been prepared from 3-piperidone phenylhydrazone derivatives. Used initially to obtain pentacyclic derivatives (35) related to the yohimbe alkaloids, this route was later extended to the synthesis of tetracyclic compounds (36). l-Methyl-5,6,7,8-tetrahydro-j8-carboline (37) was prepared in low yield by heating cyclohexanone 2-methyl-3-pyridylhydrazone with zinc chloride, a synthesis probably based on the similar preparation of the tetracyclic compound 38 starting from the corresponding quinolylhydrazine. Abramovitch and Adams extended this approach to the cyclization of cyclohexanone 3-pyri-dylhydrazone (39) itself. The main product was 6,7,8,9-tetrahydro-8-carboline (40), a smaller amount of the j8-isomer (41) also being obtained. This provides a convenient and readily reproducible route to the otherwise difficultly accessible 8-carboline ring system. A more comprehensive study on the behavior of 5-acetylenic iodides in the presence of zinc was later reported21 and clearly revealed that when iodide 14 was treated with acid-washed zinc (pre-treated with Mel for activation and removal of traces of moisture) in a mixture of benzene and DMF, the acyclic organozinc iodide 21 and the cyclic vinyl iodide 22 were both produced. Both of the above approaches employed a metal ion as a template about which the corrin cyclization was performed, but the nickel or cobalt ions could not subsequently be removed. All four isomeric selenolopyridines which can be derived from benzoselenophene (423— 426 Scheme 123) have been described. Ethyl 3-hydroxyselenolo[2,3-fe]pyridine-2-carboxy-late (429) has been prepared as shown in Scheme 124 (73BSF704). Treatment of ethyl 2-chloropyridine-3-carboxylate with methaneselenol yields (427). Nucleophilic displacement of bromine in bromoacetic acid with subsequent loss of methyl bromide yields (428), which after esterification is cyclized under Dieckmann conditions to give (429). The parent compound (423 colorless oil with b.p. 92 °C/1 mmHg) is prepared either by cyclization of compound (430) and subsequent decarboxylation of the intermediate acid (equation 57) or by reduction of 2-nitroselenophene and subsequent condensation of the amino compound with malonaldehyde bis(diethyl acetal) in the presence of zinc chloride (equation 58) (76BSF883). Selenolo[3,2-6]pyridine (426 b.p. 127-129°C/10 mmHg m.p. 35.5-37.0°C) has been obtained in an analogous manner. [Pg.1034]

Synthetically useful routes to dibenzo[c,e J[l,2]dithiins are normally based on cyclizations of biphenyI-2,2 -disulfonyl chlorides. A method applied successfully to the parent compound reduces the precursor with zinc in acetic acid to generate the bis thiol, which is then gently oxidized to the dithiin using iron(II) chloride (66HC(21-2)952). An alternative one-step reductive cyclization, which has been applied to the preparation of the 2,9- and 3,8-dinitro derivatives, involves reduction of the appropriate bis sulfonyl chlorides with hydriodic acid in acetic acid (68MI22600). Yet another reductive cyclization uses sodium sulfite followed by acidification, and these conditions lead to dibenzo[c,e][1,2]dithiin 5,5-dioxide. The first step of the reaction is reduction to the disodium salt of biphenyl-2,2 -disulfinic acid which, on acidification, forms the anhydride, i.e. dibenzo[c,e][l,2]dithiin 5,5,6-trioxide. This is not isolated, but is reduced by the medium to the 5,5-dioxide (77JOC3265). Derivatives of dibenzo[c,e] [1,2]dithiin in oxidation states other than those mentioned here are obtainable by appropriate oxidation or reduction reactions (see Section 2.26.3.1.4). [Pg.986]
