Dutt-Wormall reaction. Preparation of dia-zoaminosulfinates by reaction of diazonium salts with aryl- or alkylsulfonamides followed by alkaline hydrolysis to the corresponding sulfinic acid of the sulfonamide and the azide.

This reaction was initially reported by Dutt, Whitehead, and Wormall in 1921. It is the preparation of aromatic azide via the basic hydrolysis of aromatic diazoaminosulfinate that can be easily synthesized from aromatic diazonium salt and arylsulfonamide or alkyl-sulfonamide. Therefore, this reaction is known as the Dutt-Wormall reaction. Similarly, azides can be prepared via the cleavage of compounds from diazonium salt with ammonia, hydrazine," hydroxylamine, etc.

Hopkins and Wormall (224-226) in a series of studies have used phenyl isocyanate and p-brom phenyl isocyanate (224) on such proteins as caseinogen (casein) (224), horse serum albumin (225), and insulin (226). In studying the action of phenyl isocyanate on insulin, Hopkins and Wormall (226) observed that the amino nitrogen of the original insulin was several times greater than could be attributed to the lysine. Before they could isolate the unknown amino acid, Jensen and Evans (23) obtained the phenyl carbamido and a-naphthyl carbamido derivatives of phenylalanine from crystalline insulin treated with phenyl and a-naphthyl isocyanates. Jensen and Evans showed that, whereas there were 25 free amino groups per mole insulin before treatment with phenyl isocyanate, there were only 6 afterwards. The most recent amino acid analyses of insulin (20, 382) indicate that there are about 20 phenylalanine and only 6-8 lysine residues.
