In the first subsection of this chapter, we screened several marketed drugs, some of which contain structural moieties known to produce reactive species, for metabolic activation potential by LC/MS. Figure 10.1 shows the structures of the five test drugs, loxapine (LOX), diclofenac (DCL), qmnidine (QIN), ficlopidine (TIC) and troglitazone (TGZ). Rat liver microsomes and horseradish peroxidase (HRP) were used as the test systems with substrate concentrations of 25 or 50 /iM.
