Trazodone is predominantly metabolized by CYP3A4, which is inhibited by clarithromycin. In a double-blind crossover study in healthy volunteers, co-administration of clarithromycin increased the AUC of trazodone, prolonged its half-life, increased its C ax. and reduced its oral clearance [78 ]. There was a concomitant increase in self- and observer-rated sedation and ratings of feeling spacey , suggesting that this kinetic effect was clinically significant.
