Total synthesis of racemic

Chiral 3-alkylpiperidines were prepared through perhydropyrido[2,I-Z)][],3]benzoxazines (99TL242I, 00TA2809). 5u-Pentyl-5u,6,7,8,9,H-hexa-hydropyrido[2,I-Z ][l,3]benzoxazin-9-ones 31, 33 (R CsHn) and 34 were used in the total synthesis of racemic and natural (—)-(R) forms of adalinine alkaloid (99SL37, 99TL739). 

The addition products were used for the total synthesis of racemic aporphine, protoberine, quettamin, and phthalide alkaloids26,24. 

Nanaomycin A 103 and deoxyfrenolicin 108 are members of a group of naphthoquinone antibiotics based on the isochroman skeleton. The therapeutic potential of these natural products has attracted considerable attention, and different approaches towards their synthesis have been reported [65,66]. The formal total synthesis of racemic guanacastepene (rac-187) from Snider and co-workers (Fig. 20) was submitted six months later than the completed synthesis of Danishefsky s group [116-118]. The shortest sequence developed by the Snider group utilized the sequential cuprate addition/enolate alkylation of 2-methylcyclopent-2-enone 90 previously exploited by Piers, Williams and Danishefsky (Schemes 15 and 31). As outlined in Figs. 19 and 20, the strategies of Danishefsky and Snider are closely related. Both rely on stepwise annulations to build up the tricyclic ring system. In 1974, Wiesner and colleagues completed an elegant total synthesis of racemic napelline (34) (166). A series of model studies on this type of system was reported (167,168). They developed (169) an efficient route to the intermediate ketolactam (250) and have used this compound to complete a formal synthesis of napelline (166,170). 

The major isomer 34 is desilylated with TBAF, and oxidative cleavage of the diol with sodium periodate generates the corresponding aldehyde 35, the racemic form of which is a known intermediate in the total synthesis of racemic CP compounds. The conversion to the indoline derivative mt-31 therefore follows the strategy of the racemic route. Circular dichroism (CD) spectroscopy verified the identity of the synthetic mt-31 as the enantiomer of the naturally derived indoline (—)-31.
