Three Vehicles

The effect of iron oxide, zinc oxide and red lead on the percentage of D areas has been determined. Three vehicles were used, a pentaerythritol alkyd, a tung oil phenolic and an epoxy polyamide" . In the case of iron oxide, the D areas increased with all three vehicles in contrast zinc oxide had very little effect on the percentage D areas. However, red lead when dispersed in the alkyd and tung oil vehicles behaved in a similar way to iron oxide, whereas red lead when dispersed in the epoxypolyamide vehicle had very little effect. 

In general, the water uptake of D films tended to be higher than that of / films, but a more significant difference was shown by microhardness measurements. The results obtained with all three vehicles showed that the D areas were significantly softer than the / areas and that the distribution of the hardness values corresponded to that of the resistances. It was concluded that these films have a very heterogeneous structure and that / and D areas are brought about by differences in crosslinking density within the film. 

Temperature An examination has been made of the effect of temperature on the structural changes in polymer films produced from the three vehicles described earlier s. Three methods were used dilatometry, water absorption and ionic resistance. 

The physical form of a material destined for oral administration often presents unique challenges. Liquids can be administered as supplied or diluted with an appropriate vehicle, and powders or particulates can often be dissolved or suspended in an appropriate vehicle. However, selection of an appropriate vehicle is often difficult. Water and oil (such as the vegetable oils) are used most commonly. Materials that are not readily soluble in either water or oil can frequently be suspended in a 1% aqueous mixture of methylcellulose. Occasionally, a more concentrated methylcellulose suspension (up to 5%) may be necessary. Materials for which appropriate solutions or suspensions cannot be prepared using one of these three vehicles often present major difficulties. 

Percutaneous penetration of 7V-nitrosodiethanolamine was measured using cryo-preserved human trunk skin and three vehicle formulations (isopropyl myristate, sunscreen cream or a 10% shampoo) containing 7V-nitroso[ C]diethanolamine. The absorption rate of a low dermal dose (10 ixg/cm ) of 7V-nitrosodiethanolamine was a linear function of the concentration (0.06, 0.2 or 0.6 Xg/ xL) applied to the skin. The peak rates for the isopropyl m uistate and shampoo vehicles were seen within five hours and for the sunscreen somewhat later. Total 48-h absorption ranged from 35 to 65% of the dose and was formulation-dependent (isopropyl m uistate > shampoo > sunscreen). A total absorption of 4-6 x JcaE was estimated to equate to an applied N-nitrosodiethanolamine dose of 10 x%lcaE. When applied as a large infinite dose (0.5 mg/cm ), total 7V-nitrosodiethanolamine absorption (4-35% of the applied dose) followed a different rank order (shampoo > isopropyl m uistate > sunscreen), probably due to the barrier-damaging properties of the vehicles. The permeability coefficient for isopropyl myristate was 3.5 X 10 cm/h (Franz etal., 1993).
