Test organisms commonly used assays

Some important assays commonly used in biochemical genetics laboratories do not provide quantitative data (e.g. MPS-EP, qualitative urinary organic acid analysis, AA-TLC). In addition, all successful investigations depend heavily upon selection of the correct analytes to measure and the appropriate interpretation of the quantitative or qualitative results in their clinical context. These challenges suggest a requirement for external quality assessment or proficiency testing schemes that can inform participants about their performance in these areas when compared with other centres. 

Waksman and Reilly have summarized the factors which have a bearing upon the choice of the method to be employed in measuring quantitatively the activity or potency of an antibiotic substance. They have cited literature references to the more commonly used methods. Loo and coworkers have described a suitable method for the quantitative determination of streptomycin by the filter paper disc, agar plate diffusion technique using Bacillus subtilis as the test organism. The procedure proved satisfactory in the assay of surface and submerged culture beers and of preparations obtained in isolation and purification... 

However, despite the common use of these assays, only a small fraction of wild organisms actually grow in commercially available artificial media. For example, estimates for SRB in marine sediments indicate that only one in a thousand of the organisms present actually show up in standard growth tests [22].
