Tartrate resistant acid phosphatase TRAP

Bone resorption markers are Listed in Box 49-5. N- and C-telopeptides of type I collagen (NTx and CTx) and deoxypyridinoline are most frequently measured. Other markers include serum tartrate-resistant acid phosphatase (TRAP) and hydroxyproline. Bone resorption markers, apart from TRAP, were initially measured in urine. More recently, methods using serum have been developed for a number of these analytes including N- and C-telopeptides. 

To count multinucleated cells, cultured cells were stained as described below. After cells were cultured, adherent cells were fixed with 10% formaldehyde in phosphate-buffered saline (—) for 20 min. After cells were treated with 95% ethanol for 1 min, the well surface was dried and treated with tartrate-resistant acid phosphatase (TRAP)-staining solution [0.1 M sodium acetate buffer (pH 5.0) containing 50 mM sodium tartrate, 0.1 mg/ ml naphthol AS-MX phosphate (Sigma Chemical Co., St. Louis, USA), and 1 mg/ml fast red violet LB salt (Sigma Chemical Co.)] for 30 min. TRAP-(-l-) multinucleated cells were then counted under a microscope.

Bone In 72 kidney transplant patients on TAC for more than 5 months (5-51 months), 27.78% of patients had bone loss and the loss rates were statistically different between the high TAC group (TAC>6ng/mL) and low TAC group (TAC<6ng/mL) (45.5% vs 20.0%, p=0.026). Correlation analysis showed that TAC concentrations were positively correlated with tartrate-resistant acid phosphatase-5b (TRAP-5b) in male recipients (r=0.287, p< 0.05) [57 ].
