Targeting assessment purpose

Several research groups have demonstrated that species differences in the biotransformation of drugs with diverse but known in vivo metabolite patterns, could be reproduced with hepatocytes. Hepatocytes from diverse species like rats, rabbits, dogs, monkeys and man have been used to study the biotransformation of tolbutamide 37), amphetamine 38), ketitofen 39) and diazepam 40). These and other comparative studies again demonstrate that in general extrapolation of data obtained with cellular fractions, hepatocytes or intact animals from one species to another species is not possible, and stresses the need for the use of target-animals for risk-assessment purposes. [Pg.75]

CMs include AChE inhibition, particularly in the blood and brain behavioral and/or motor activity changes and developmental toxicity or developmental neurotoxicity. When EPA evaluates AChE data for risk assessment purposes, measurements of AChE from the target tis.sue — the central or peripheral nervous system — are preferred. Toxicology studies submitted to EPA for pesticide registration typically measure plasma, red blood cell (RBC), and brain cholinesterase (ChE) inhibition and most often do not measure peripheral tissues. In the absence of such data, EPA considers blood measures as surrogate.s of peripheral tissues,... [Pg.619]

The matrices to be validated depend on the target/purpose of the study, e.g., blood, urine, muscle, or liver. The latter two may be covered by methods developed for food of animal origin. The method must take into account all relevant compounds used in the assessment of risk to consumers/operators or bystanders. The required LOQ... [Pg.35]

The third criterion is that the model should target an endpoint relevant for REACH. Only models that address the endpoints of interest for REACH are appropriate within this purpose. We notice that REACH mentions different purposes for the QSAR models classification and labeling, is one possible target of the model, and risk assessment in another. In the first case models are classifiers in the second case a regression more is more suitable. Indeed, in the first case the... [Pg.85]

Amino acid analysis has long been of importance for nutritional purposes. Recently, there has been an increasing interest in assessing the exact amino acid composition of new protein sources, infant formulas, nutraceutical-type products, or supplements targeted at people with unusual metabolic needs. There is also a regulatory necessity to have rapid methods available for amino acid determination to verify product consistency with that declared on the label [196]. [Pg.582]

Process simulation units shall not be required to be inverted at some point during the incubation period. All hlled units shall be sufficiently manipulated to assure the contact of all sterile surfaces by the growth media prior to incubation. Momentary inversion of test units shall be surfaces of the container/closure system. Reconciliation requirements of process simulation units shall be equivalent to the requirement for production. The target will be 100% reconciliation/accountability of all hlled units. Any excursion must be investigated and documented however, a variance is not an automatic invalidation of a process simulation test. Process simulation testing shall simulate normal production as closely as possible because its purpose is to assess the potential of contamination in units representative of normal production. [Pg.907]

Accuracy, in general, is assessed using a minimum of nine determinations over a minimum of three concentration levels covering a range of 50-150% of the target concentration (e.g., three concentration levels with three replicates each). The criteria for acceptable accuracy of the assay results cannot be generalized because accuracy depends on the concentrations of the analytes being evaluated and the acceptable criteria depend on the purpose of the analysis (23). [Pg.754]
