We have used the nitration of a tyrosine analog with superoxide dismutase to measure the peroxynitrite production from activated rat alveolar macrophages (Fig. 38). The estimated rate of peroxynitrite synthesis was estimated to be 0.1 nmol/10 cells/min (Ischiropoulos et al., 1992a). The rate of nitration was the same whether native Cu,Zn-superoxide dismutase or the phenylglyoxyl-H202 modified superoxide dismutase (which is >99% inhibited with respect to its superoxide scavenging activity) was used (Fig. 39). Three other independent but indirect estimates of peroxynitrite formation were consistent with the superoxide... [Pg.64]

Mulberroside F (104) showed moderate inhibitory effects on tyrosinase activity and on the melanin formation of melan-a cells. It also exhibited superoxide scavenging activity that is involved in the protection against auto-oxidation [62]. The geranylated stilbenes 81 and chlorophorin from Chlorophora excelsa [51], prenylated ones 4-prenyloxyresveratrol and artocarbene (72) from Artocarpus incisus [528] all demonstrated tyrosinase inhibitory activity (ICso 96, 1.3, 0.66 and 2.45 //M, respectively). [Pg.610]

Cu(TpMe)2] prepared from the reaction of CuCl2 with K(TpMe) has been characterized by magnetic susceptibilities, fast atom bombardment and IR spectrometry, and X-ray single-crystal study.1 Cop-per(I) complexes containing water soluble phosphines, [Cu(TpMe)(L)] (L = 4- or 2-(diphenylphosphane)benzoic acid), have been characterized by IR in the solid state, and by NMR (JH and 31P 1H ) and electrospray mass spectroscopy in solution. Chemiluminescence technique was used to evaluate their superoxide scavenging activity.2... [Pg.275]

Compounds 25, 26 and 27 belonging to I-C group, isolated from Vatica rassak and V. indica, showed antioxidation and SOD like activities, indicating that this group of oligomers also has antioxidation activity and superoxide scavenging activity [58,59, 60]. [Pg.622]

Chemiluminescent assay for superoxide-scavenging activity of aldehyde-modified HSA... [Pg.40]

Xanthine/xanthine oxidase (X/XO) system was used to generate O2 to measure the superoxide-scavenging activity of aldehyde-modified HSA. Briefly, 1 ml solution contained 100 pg/ml X, 0.01 U/ml XO, 250 pM DTPA and 500 pM luminol in the absence or presence of 10 nM of aldehyde-modifed HSA. Chemiluminescent intensity was recorded as described above. [Pg.40]
