Sumatriptan dosage

Naratriptan is also dosed at 2.5 mg. Maximum dosage is 5 mg/d. Relief is achieved in 4 hours in 65% of the patients. The rate of reeurrence, an estimated 27%, is similar to that for sumatriptan and zolmitriptan. Frovatriptan is also dosed at 2.5 mg. Relief is achieved in 2 hours in 42% of the patients. Eletriptan is dosed at 40 mg with a maximum dosage at 80 mg. Headache response is achieved in 2 hours in 60% of the patients. Recurence rate is estimated at about 20%. Rizatriptan is dosed at 5 mg with a maximum dosage at 10 mg. Headache response is achieved in 2 hours in 62% of the patients. Recurence rate is estimated at about 39%. 

Studies in rabbits and mice showed that oral and intravenous doses of 100 mg/kg/ day and 2 mg/kg/day of sumatriptan, respectively, to be lethal to embryos, whereas dosages as low as 2 mg sumatriptan/kg/day over a 1-month period led to comeal opacities and defects in the cornea epithelium in dogs (GSK, 2006). No controlled studies have been conducted to investigate this aspect in humans. 

A 28-year-old man with migraine and vascular pain of the face was given verapamil 480 mg/day when sumatriptan did not control the pain. Verapamil was withdrawn after 1 month and reintroduced in a dosage of 960 mg/day after a new episode of pain. He then developed arthralgia in the right hand and arm, which disappeared after withdrawal and recurred after rechallenge. 

Although the information is limited, it shows that no sumatriptan or zolmitriptan dosage adjustments are expected to be needed if used with pizotifen. On the basis of the information on sumatriptan and zolmitriptan it seems unlikely that any of the other triptans will interact. 

Sumatriptan can he found as the free base but is most commonly prepared as the succinate salt. Dosages depend on the route of administration.
