Strategic themes

The final route, which has ultimately become the manufacturing process for aprepitant, utilized many of the same concepts as the previously discussed routes, but there was a significant strategic difference. The novel aspect of the plan, from a retrosynthetic analysis perspective, was that the stereocontrol element was chiral alcohol 37 and not one of the two stereocenters contained in the morpholine backbone The strategic themes that were conserved from some of the other routes included an acetalization and resolution at an epimerizable center. ... 

Porter s framework begins with strategic themes. Strategic themes are the cornerstones of the activity system. Porter observes that the themes require clear choices regarding how to compete. This is a difficult but necessary step and one that should not be taken lightly. Too often, companies try to be all things to all people. Failure to choose how one will or will not compete means there is no strategy at all. 

With strategic themes in place. Acme had to develop supporting activities and processes that implemented those themes. Figure 10.2 shows some of the activities Acme might have pursued to implement the strategic themes. The figure shows the links between the activities and themes by way of lines. Some activities support not just themes, but also other activities. 

First order fit between the activity and strategic theme. In the activity map, activity costing fits the notion of measuring contribution on different pieces of the business. Its application is as an internal control to evaluate product and customer profitability. Product R D is another example of first-order fit. Intramolecular Heck reactions for building up complex oxacyclic skeletons are a common theme in the synthesis of natural products. These reactions are exceptionally valuable for the installation of quaternary carbon stereocenters. In the morphine total syntheses by Overman <1994PAC1423> and Trost et al., intramolecular Heck reactions to form dihydrobenzofurans served as strategic key steps (Equation 138) <2005JA14785>. Asymmetric variants of intramolecular Heck reactions based on BINAP ligands to yield dihydrobenzofurans have also been investigated <1998T4579>.
