Storage of cells

The long-term storage of cells at low temperature is now routine. Methods awaited the observation that glycerol exerts a marked protective effect on the cells during the freezing process. 

In order to use a programmable freezer it is essential to have a supply of piped liquid nitrogen from a pressurised cylinder and this is also very convenient for topping up liquid nitrogen freezers. 

Complete 96 well microtitre plates and their contents can also be frozen in a programmable freezer. The medium in the wells should be replaced with freezing medium ( 7.3.1) and the whole plate cooled on ice and taped shut and wrapped in plastic film prior to freezing.

Blood and lymph are approximately isotonic to a cell so that cells do not gain or lose liquid when bathed in these fluids. Pure water is hypotonic and may cause cells to swell and burst. During intravenous feeding, injections, and storage of cell tissue, a salt (saline) solution is used with a concentration of solutes that is essentially isotonic with blood (and hence, with the cell) to prevent cell damage. 

Ciyopreservation Ultra-low temperature storage of cells, tissues, embryos or seeds. This storage is usually carried out using temperatures below -100°C.
