Staining controls

As a test of reproducibility of spot printing and staining, a 10 spot by 10 row array (100 spots total) were printed on three slides. All slides were stained in a single staining run, using the same secondary detection chemistry with DAB as a chromogen. While DAB cannot provide accurate photometric measurements, it can provide relative comparisons from one spot to another on the same slide. When the stained control slides were analyzed in this... [Pg.181]

Fig. 10.19 Lack of toxic effects of CM fullerene on breast epithelial cells. Cm does not inhibit cell proliferation. MCF 10A and (A) MDA MB 231 (B) breast cancer cell lines were cultured either in the absence or presence of methanol C60 and cell proliferation was assayed by crystal violet staining. Control, no C 1 Opg Cm, A 50 pg Cm, X 250 pg CM. (C). MDA MB 231 cells were simultaneously stained with calcein and ethidium using a live-dead assay kit. Lack of red-colored cells and the presence of cells stained in green indicate the lack of toxicity (D). MDA MB 231 cells were either untreated (open box) cultured with varying amounts 10 (gray ), 50 (patterned ) and lOOpg (filled ) of C60 for 48 h and analyzed for cell cycle progression by flow cytometry (Levi et al., 2006) (See Color Plates)... Controls are used to ensure proper technique and the specificity of the stain. The use of similarly processed positive and negative staining controls is essential for interpretation of immunohistochemical reactions and must he done for each anti-hody. These controls may he commercially prepared, previously tested patient samples or proficiency testing specimens for which results have heen confirmed. [Pg.408]

Scale and Stain Controllers, Polyacrylates (low molecular weight) and organic phosphonates, eg, (l-hydroxyethyHdene)diphosphonic acid, prevent or control precipitation of CaCO by acting as chelating agents (qv) or dispersants (qv) to prevent excessive formation of hard scale by promoting crystal distortion. [Pg.302]

The value for zero stain control is subtracted from the values obtained for all wells, and the corrected density is then plotted as a percentage of the zero toxicity control (Fig. 2). The definitive value for toxicity is that at which stain uptake is reduced by 50% (the TC50). [Pg.125]

Split each cell suspension equally into two tubes (50 pL in each), one for the non-stained control and one for the staining of CD3" T cells. [Pg.258]

In order to minimize spectral overlap during a multicolor flow cytometry analysis, a compensation procedure before acquiring data is required. Use the FACSDiva software package to automatically calculate a compensation matrix from single-stained controls (CompBeads, BD immunocytometry system, 552843 see Table 4). Below, a brief guide for getting started ... [Pg.450]
