Sparteine sulfate

Sparteine is liberated from the commercially available sulfate salt (Aldrich Chemical Company, Inc.) as follows Sparteine sulfate pentahydrate (100 g, 240 mmol) is dissolved in deionized water (125 mL), and to this solution is slowly added aqueous 20% NaOH (100 mL). The resulting milky-white, oily mixture is then extracted with ether (4 x 150 mL). The combined ethereal extracts are dried over anhydrous K2CO3, filtered, and the solvent is removed under reduced pressure. Vacuum distillation of the residual oil from CaH2 affords 52 g (92%) of sparteine as a clear, colorless to slightly yellow, viscous oil (bp 115-120°C/0.3 mm). The sparteine free base readily absorbs atmospheric carbon dioxide (CO2) and should be stored under argon at -20°C in a freezer.

Two C16H26N 2 compounds were also isolated from sparteine sulfate mother liquors as obtained in the purification of the plant extracts (271) which are apparently structural isomers of XCI. These compounds, [ ]d — 13.58° and 120.40° in ethanol, gave very strong pine splinter tests for the pyrrole ring. Moureu and Valeur (251) postulated that their isosparteine, [aj 25.0° in ethanol (picrate, m.p. 178°), obtained by a Hofmann degradation and reversal, also contained a methylpyr-rolidine ring. 

Patients given thiamyiai sodium had a marked increase in cardiac arrhythmias when they were given intravenous sparteine sulfate, but those given thiopentai or etomidate did not.

Sparteine sulfate possesses several important biological activities, such as improving the tuning failure (a rhythmic disorder) of the heart, and produces a unique and periodic oxytocic action [2]. Consequently, as an injection, it is applied in cases of tachycardia, arrhythmia, uterine contraction disorder, inertia uteri, etc. 

The addition of lupin seeds to the food of diabetic-hypercholesterolemic rabbits decreases cholesterol levels and postprandial hyperglycemia. Sparteine sulfate administered by intravenous infusirai to normal men increases either basal or glucose-induced secretion sparteine administration to patients with type 2 diabetes causes a fall in plasma glucose levels [56]. 

The most common tetracyclic alkaloid, (-)-sparteine, was isolated from Cytisus scoparius with (-)-isosparteine and (+)-lupanine. Sparteine sulfate has been used as an oxytocic.
