Solasodine glycoside

We present, here, a simple determination method for solasodine glycosides by using thin-layer chromatography (TLC)-immunostaining. 

Solasodine glycosides were applied to TLC plates and developed with chloroform-methanol-ammonia solution (7 2.5 1). A developed TLC plate was dried and then sprayed with blotting solution mixture of isopropanol-methanol-water (5 20 40, by volume). It was placed on a stainless-steel plate, then covered with a PVDF membrane sheet. After covering with a glass microfiber filter sheet, the whole plate was pressed evenly for 45 s with a 130°C iron, as previously described [11], but with a modification. The PVDF membrane was separated from the plate and dried.

The alkaloid contents of 5. xanthocarpum collected in Nepal and France have been compared. Fruits from the French plant were richer in solasodine glycosides than those from the Nepalese plant (yields of solasodine 4.6% and 1.6% respectively). The French plant, in addition, contained traces of tomatidenol glycosides in the stalks. A number of Solarium species have been investigated for their aglycone content. After solasodine glycosides were transfered to the PVDF membrane sheet from the TLC plate by heating as previously reported,the PVDF membrane was treated with NaI04 solution, followed by conjugation with BSA, because solasodine glycosides on PVDF membrane are washed out by buffer solution or water without the formation of conjugate with carrier protein. The PVDF membrane was immersed in antisolamargine MAb and then peroxidase-labeled secondary MAb. When the substrate and were added, clear blue spots appeared. 

Other solasodine glycosides are solasonine (with L-rhamnose and D- ucose), and a-solamargine, the first glycoside isolated from Solanum marginatum (with two molecules of L-rhamnose and one molecule D-glucose). Solasodine is used as starting material for the laboratory synthesis of steroid hormones. P-Solasonine see a-Solasonine. 

In addition to solaradixine, the fruit of 5. mammosum contains two other solasodine glycosides, the main product of which was shown to be a trioside consisting of 2 mol of D-glucose and 1 mol of L-rhamnose (95). 

The distribution of solasodine glycosides in leaves, flowers, and fruits of S. laciniatum has been studied by Weiler et al. 371) using radioimmunoassay. The highest levels of alkaloids are found in empty anthers and in the inner pericarp of green fruits.
