Because excited triplet states decay more slowly than excited singlet states, it is much easier to determine the excited triplet-state lifetime 3t than H. Phosphorescence emission from a degassed sample at low temperature (77K) lasts for longer than 1ms and may even be several seconds. The molecules in the sample are irradiated with a short ( 1 ps) flash and the decay of the phosphorescence signal is monitored using an oscilloscope. Any accompanying fluorescence signal will decay too rapidly to be observed. The excited triplet-state lifetime is obtained as the time taken for the emission intensity to fall to 1/e of its initial value. [Pg.58]

By this time it was demonstrated that the [3Fe-4S]W+ form of aconitase is inactive, while the [4Fe-4S]2+ form is active. How is the activity of the enzyme affected by the oxidation state of the [4Fe-4S] cluster Because the active enzyme contains a [4Fe-4S]2+ cluster, either the 3+ or 1+ oxidation states may also be stable. The 3+ state is unstable since oxidation of the [4Fe-4S]2+ resulted in the immediate loss of a ferrous ion and conversion to a [3Fe-4S]i+ cluster (46,47). However, reduction of active aconitase by sodium dithionite or photoreduction in the presence of deazaflavin produced in high yields an EPR signal characteristic for [4Fe-4S]l+ clusters (47). When active enzyme within an anaerobic assay cuvette was photoreduced, the activity of the enzyme dropped to 1/3 of its initial value. Further photoreduction resulted in cluster destruction. Then, if the enzyme is reoxidized with air, the activity returned to its original value. This demonstrated that the redox state of the cluster can modulate the enzyme activity. Use the same series of data and follow the same procedure as before to try out the Lorentz-Gauss convert window type. There is one single parameter LB available to adjust the window. Set the initial value to LB = 0, increment and decrement its value in small steps and inspect the shape of the window using the interactive mode. Note that for LB > 0 the shape of the window is similar to the exponential window (signal-to-noise improvement) whereas for LB < 0 the window shape is similar to the sine-bell squared window. The modified electrode showed a very good stability even at pH 9. A phase-locked loop is a bit of a hybrid of the oscillator circuit and the propagation measurement system discussed in the previous two sections.
