Serum Total Iron and Total Iron Binding Capacity The results for the determination of serum total iron and total iron-binding capacity of rats fed treatment diets for 18 months are also listed in Table VI. A significant increase in serum total iron was detected in rats fed the Maillard browned egg albumin over their control group. Increased serum total iron with normal total iron binding capacity is associated with hemolytic anemia, hemochromatosis, hemosiderosis, and hepatitis (30 ). The effect of administration of benzene on iron metabolism was investigated by giving 0.5 mL/kg intraperitoneally or 1 mL/kg subcutaneously to female rats for 10 consecutive days (Ahmad et al. 1994). Serum from benzene-treated female rats offered less protection against iron-catalyzed lipid peroxidation of brain tissue than the serum of control female rats. Serum iron concentration and total iron binding capacity did not exhibit any significant change after benzene administration. Iron saturation of transferritin was not affected by benzene. [Pg.205]

McPherson and George 89) determined total copper and zinc of red cells and the free copper and zinc of plasma and dialysis fluids of patients undergoing regular hemodialysis, using atomic absorption spectroscopy. Immunoprecipitation of ferritin from heart tissue extracts. The remaining suspension of homogenized heart tissue in the lysis buffer was incubated at 70°C for 10 min, cooled on ice and centrifuged at 10,000 g for 20 min. The supernatant was collected and the pellet discarded. Total amount of ferritin in each extract was calculated using the results of the ELISA. Saturating amounts of immune serum against rat heart ferritin were then added to the extracts. The mixtures were incubated at 4°C for 72 h, immunoprecipitates were washed twice in lysis buffer and stored dry at -80°C until analyzed for iron concentration by atomic absorbtion. [Pg.50]
