Selective sequencing

It should be noted that in almost all cases only one fold exists for any given sequence. The uniqueness of the native state arises from the fact that the interactions that stabilize the native strucmre significantly destabilize alternate folds of the same amino acid sequence. That is, evolution has selected sequences with a deep energy minimum for the native state, thus eliminating misfolded or partly unfolded structures at physiological temperatures. [Pg.372]

Early diffraction photographs of such DNA fibers taken by Rosalind Franklin and Maurice Wilkins in London and interpreted by James Watson and Francis Crick in Cambridge revealed two types of DNA structures A-DNA and B-DNA. The B-DNA form is obtained when DNA is fully hydrated as it is in vivo. A-DNA is obtained under dehydrated nonphysiological conditions. Improvements in the methods for the chemical synthesis of DNA have recently made it possible to study crystals of short DNA molecules of any selected sequence. The presence of hydroxyaluminum- and hydroxyaluminosilicate polymer in interlayered montmorillonite greatly promotes the adsorption of Cd, Zn, and Pb (Saha et al., 2001). The adsorption selectivity sequences of montmorillonite (Pb > Zn > Cd) and interlayered montmorillonite (Pb Zn Cd) resemble the metal selectivity on amorphous Fe and Al hydroxides (Saha et al., 2001). Greater adsorption of trace metals is found at higher pH and C02(g) concentrations. Sites available for Zn2+ sorption are less than 10% of the Ca2+ sites on the calcite surface, and Zn adsorption is independent of surface charge. This indicates a surface complex with a covalent character (Zachara et al., 1991). Furthermore, the surface complex remains hydrated and labile because Zn2+ is rapidly exchangeable with Ca2+, Zn2+ and ZnOH. At the dolomite-solution interface, the carbonate(C03)-metal (Ca/Mg) complex dominates surface speciation at pH > 8, but at pH 4-8, hydroxide (OH) -metal (Ca/Mg) dominates surface speciation (Pokrovsky et al., 1999). Calcite has an observed selectivity sequence Cd > Zn > Mn > Co > Ni > Ba = Sr, but their sorption reversibility is correlated with the hydration energies of the metal sorbates. Cadmium and Mn dehydrate soon after adsorption to calcite and form a precipitate, while Zn, Co and Ni form surface complexes, remaining hydrated until the ions are incorporated into the structure by recystallization (Zachara et al., 1991). [Pg.148]

In glycerol monooleate/decane bilayers we find the steady-state conductance at zero current to be proportional to the first power of the ion concentration and to the second power of the ionophore concentration, as illustrated in Fig. 1.
