Selection of clones

For expression of BCCP-p53 fusion proteins, we typically found the protein concentration in crude lysates to be 5 mg/mL, and we estimated that BCCP-p53 was present at approx 1% of total soluble protein. When expressing a number of clones in parallel for array fabrication, the Bradford assay can conveniently be done on all clones in parallel using a microtiter plate format. However, it would be laborious to carry out SDS-PAGE analysis on all clones, so typically we assess only a selection of clones in this way, since the absolute expression level is not critical for array fabrication.

The objective of phage display technology is the selection of clones of interest from a library of mutants. The diversity of the library will be the number of individual clones present in the collection whereas its quality will be related to the method used for its creation and to the functional diversity, i.e. the diversity from which all the clones that are unstable, misfolded or degraded have been removed. A good library will feature a high diversity and a high quality. 

The oils produced by a range of different clones grown in Malaysia have been analyzed [95]. The clones were derived from early tissue culture experiments and were not from selected palms. While there was a wide variation in oil composition among the clones, there was a high within-clone uniformity, indicating a strong genetic control of oil composition. Thus, selection of clones for oil quality should be possible. 

Because virus diseases, in particular fanleaf, have a major impact on vine performance under the cooler German conditions, visual assessment and performance-based selection of clones gave good results. When viruses as causal agents of vine decline...
