There are two types of scanning acoustic microscopes. If the illumination and the reception of the acoustic waves are performed by two identical lenses arranged confocally, the SAM is called a transmission SAM. The lens geometry used for transmission imaging is shown schematically in Fig. 41 [93]. [Pg.28]

Figure 12. Schematic diagram of the scanning laser confocal microscope. The out-of-focus information that normally reaches the eyepiece (detector) and leads to difficulties in interpreting optical images is rejected because the optical path does not take it through the pinhole. By scanning the incident laser beam across the sample, a digitized image is constructed from the infocus light rays that pass through the pinhole. Figure 12. The more comonly applied confocal laser scanning microscope (CLSM) features a variable pinhole and allows adjustment of magnification by modifying scan angles. A schematic example of the principle of confocal laser scanning is given in... [Pg.1071]

Figure 6.1. Schematic diagram of a laser scanning coirfocal microscope. The critical confocal feature is the pinhole aperture in front of the detector that rejects light coming from outside the focal plane. The transmitted light mode shown is not confocal. Figure 6.1. Fig. 6.1 Schematic of a laser confocal scanning microscope. The laser illumination spot is scanned across the sample. Reflected light is de-scanned and passed through the beam splitter and the confocal aperture to the detector. The detector of transmitted light may be present as an accessory, but it gives a normal, not a confocal, optical microscope image. Fig. 6.1 Schematic of a laser confocal scanning microscope.
