MAHMA NONOate spontaneously dissociates in a pH-dependent, first-order process with a half-life of 1 min at 37 °C. In vivo, examination in anesthetised rats showed, that MAHMA NONOate had both platelet inhibitory and vasodepressor effects [50]. Like other NONOates, MAHMA NONOate inhibited collagen-induced and ADP-induced rat platelet aggregation in a concentration-dependent manner [51]. ODQ, a soluble GS inhibitor, caused only small influence on the concentration-response curve to MAHMA NONOate, indicating that cGMP-independent mechanisms play a crucial role. A potential target of MAHMA NONOate induced inhibition was the sarco-endoplasmic reticulum calcium-ATPase of the platelet dense tubular system... [Pg.242]

The molecules of all the apically and ribbed-carboxymethylated cobalt(III) sarcophaginates (N-protonated, neutral, and carboxy-deprotonated, and the nitrozation product) as well as their hexacoordinate 1,8-bis- and 1,3,8-tris-carboxymethylated analogs adopt the leh configuration [117, 232]. The chirality of the coordination polymeric cobalt(III) l,8-bis(carboxymethylamino)sarco-phaginate / Mn2+, Mn22+, Co +, and Zn + systems was studied by X-ray crystallography [233]. [Pg.149]

Expression of Ion Channels and Pumps Differences in the expression of voltage-gated ion channels at the sarco-lemma, T-tubular system, or SR may determine the excitation of a muscle fiber in response to neurotransmitter release. A decrease in the amount of T-tubules measured per unit volume or voltage-gated Na channels would potentially act to decrease the amplitude of the muscle action potential. Similarly, Ca entry and release from intracellular stores (mainly the SR) influence thin filament activation and, ultimately, force generation. [Pg.1093]
