Finally, also isolated from D. vulgaris was another homodimeric protein (subunit molecular mass of 27 kDa) named nigerythrin (43). Spectroscopic characterization revealed that this protein is similar to Rr. The function of nigerythrin is also unknown. It should be noted that genes coding for two similar Rr proteins were also found in Arch-aeoglobus fulgidus.

In the plantaricin system, five operons are coordinately controlled by the plantaricin A signaling system. In addition to the regulatory operon, two operons encode bacteriocin precursors and immunity proteins, one operon encodes the transport system of the bacteriocins and the PP itself, and the last operon contains four genes of unknown function (it could be a bacteriocin encoding operon but no activity has been found so far). The key operon encoding the three-component signaling pathway (composed of a PP, an HPK, and two RRs) will be discussed below.

Many destination vectors are commercially available. However, it is also possible to construct a destination vector that is suitable for individual experiments. To constrnct snch vectors, one should insert the rr ffi-containg cassette (commercially available from Invitrogen) into the appropriate locns of a Gateway-incompatible vector. The resultant plasmids can be amplified only in the DB3.I strain, due to the toxic ccd gene (7).
