With a high theoretical plate number of a column you can expect sharp peaks. With a given selectivity, resolution is improved by sharp peaks since the resolution is defined as the distance between two peaks at the peak base and not peak maximum. Resolution is the ability to distinguish between closely spaced peaks in a spectrum. The resolution is given as the value of the full width of a peak at half of its maximum height (FWHM). This value is given in keV for a selected full-energy peak, usually for the 1332 keV peak of cobalt-60. Sometimes resolution is defined as a percentage of the absolute peak position. The shape of a peak and also the resolution is influenced by the count rate. [Pg.4191]

RCs were isolated from Rb. sphaeroides w.t. by anion exchange chromatography. The measurements were carried out by the time-correlated single-photon-counting technique as described. The full width at half maximum of the system temporal response function was about 30 ps. The positron lifetime experiments were carried out with a fast-slow coincidence ORTEC system with a time resolution of about 230 ps full width at half maximum. A 5mCi source of Na was sandwiched between two identical samples, and the total count was one million.
