In striatal neurons, the effects of D2 receptor activation on K+ channels are more complex. Stimulation of the D2 receptor has been reported to open a K+ channel that displays a 85 pS conductance and a weak inward rectification, and this rectification seems to differ from that found in pituitary cells (Freedman and Weight, 1988 Einhorn et al., 1991 Greif et al., 1995). In contrast, D2 receptor activation was also reported to suppress K+ currents probably through Kir2 channels (Uchimura and North, 1990). However, the activation of K+ channels occurs in a membrane-delimited manner via Gpy mobilization, whereas inhibition of K+ channels could result from the D2 receptor mediated inhibition of adenylyl cyclase and the dephosphorylation of Kir2 subunit at its PKA sensitive site (Nicola et al., 2000). [Pg.125]

To detect structural variations in the O-PS structure of A. salmonicida from typical and atypical isolates we have used a microscale CE—MS-based method for analysis of A. salmonicida LPS directly on the bacterial cells. This method involves pretreatment ofbacterial cells ( 10 —10lu cells) with proteinase K, RNase and DNase followed by delipidation with mild acetic acid and subsequent MS analysis. Using this method, we have analyzed LPS structure of 39 typical and atypical isolates of A. salmonicida and related species (Table 1). Calamitic metallomesogens forming a chiral smectic C phase (SmC ) are ferroelectric materials. Due to the low symmetry of this phase when the helix is unwound (C2) the molecular dipoles are aUgned within the layers of the SmC phase, giving rise to ferroelectric order in the layers. Because the SmC phase has a helical structure, there is no net macroscopic dipole moment for the bulk phase. However, it is possible to unwind the helix by application of an external electric field or by surface anchoring in thin cells. The synthesis of PS and transport to the mitochondria have been successfully reconstituted using permeabilized cells [25]. The transport of PS to the mitochondria in per-meabilized mammalian cells occurs in the absence of cytosol, displays an absolute requirement for ATP, and occurs with a t,/2 of approximately 3 h at 37°C. This transport does not require ongoing synthesis of PS, and 45-fold dilution of the permeabilized cells does not alter the rate or extent of transport. These results are consistent with a membrane-bound transport intermediate that utilizes zones of close membrane apposition between the ER and the mitochondria. Although there is no absolute requirement for cytosol in the transport reaction, a soluble 9-kDa Ca -binding protein, named SIOOB, that is highly conserved across mammalian species can enhance the transport several fold (O. Kuge, 2001). Permeabilized yeast have also been used to examine PS transport (G. Achleitner, 1995). Unlike mammalian cells, the transport of PS to yeast mitochondria does not require ATP. [Pg.467]
