Starting from fresh tissues, or tissues dehydrated with acetone, a limited amount of the mucopolysaccharide can be extracted with water or with salt solution. However, a large proportion seems to be bound to protein, and solubilization requires degradation of the protein part with enz mnes, or scission of the carbohydrate-protein linkage with weak alkali. 

Glycoproteins or mucoproieins. Compounds of proteins with carbohydrates. All glycoproteins contain a hexosamine and usually sulphate, ethanoate and glucuronic acid. The carbohydrate-protein linkages are, in some cases covalent and in others of the salt type. Aqueous solutions of glycoproteins are extremely viscous.

AMCA-NHS, succinimidyl-7-amino-4-methylcoumarin-3-acetic acid, is an amine-reactive derivative of AMCA containing an NHS ester on its carboxylate group (Thermo Fisher). The result is reactivity directed toward amine-containing molecules, forming amide linkages with the AMCA fluorophore (Figure 9.23). Proteins labeled with AMCA show little-to-no effect on the isoelectric point of the molecule.
