Proprietary screening methods

After screening about 10000 individual variant clones, more than 60 primary hit clones with more than 50% increased stability were selected and recombined using a proprietary recombination method [28]. After two rounds of recombination and screening with increased CIAA concentrations, ten clones were selected for retesting on shake-flask scale. 

Screening of proprietary libraries provides a unique opportunity for the identification of novel scaffolds. Such libraries contain either isolated natural products or complex samples based on fermentation broths. These libraries can be screened in biological assays to identify active molecules. Acrylonitrile is commercially produced from propylene by a molybdate-based catalyst that has been optimized to produce a yield of around 80% acrylonitrile. Utilizing a less-expensive feedstock, the selective ammoxidation of propane to acrylonitrile has significant potential in reducing acrylonitrile production cost. The work-flow for this chemistry consisted of a primary scale evaporative synthesis station and 256-channel parallel screening reactor using a proprietary optical-based detection method. Access to a good proprietary collection of enzymes is also extremely helpful for finding an enzyme, especially if those collections have been logically assembled for applications of interest.
