Preliminary tests of the system

If the pore size of the support is selected so that molecules M can partially penetrate into it, a deviation from the Gaussian would indicate an association equilibria of type M + M = M2, for example. This association mey or may not be ligand induced. The analysis of the peak shape of rapidly reversible equilibria has often been attempted, but it has usually been found to be of little value compared to the equilibrium saturation techniques where reactions between macromolecules and small ligands are concerned 

In the case of slowly equilibrating systems, the static exclusion methods are obviously preferable to the dynamic ones. Deviations of actual elution volumes of reactants fron the presumed ones may indicate the adsorption effects of the support.
