Voriconazole Prednisolone

Drugs affected by voriconazole include the following benzodiazepines, calcium channel blockers, cisapride, coumarin anticoagulants, cyclosporine, ergot alkaloids, HMG-CoA reductase inhibitors, NNRTIs, phenytoin, protease inhibitors, pimozide, proton pump inhibitors, quinidine, prednisolone, rifabutin, sirolimus, sulfonylureas, tacrolimus, vinca alkaloids.

Clinically important, potentially hazardous interactions with acetylcysteine, adenosine, aprepitant, aripiprazole, buprenorphine, caffeine, charcoal, clarithromycin, clobazam, dorazepate, clozapine, darunavir, dasatinib, delavirdine, dexamethasone, diltiazem, doxacurium, erythromycin, felodipine, fesoterodine, fosamprenavir, imatinib, influenza vaccines, lacosamide, lapatinib, levetiracetam, lopinavir, methylprednisolone, midazolam, nelfinavir, nilotinib, piracetam, prednisolone, propoxyphene, ritonavir, rivaroxaban, rufinamide, solifenacin, St John s wort, telithromycin, temsirolimus, terbinafine, tolvaptan, troleandomycin, verapamil, voriconazole... 

Voriconazole increases plasma levels of prednisolone but not to a clinically significant extent A case report notes that voriconazole appears not to interact with oral hydrocortisone. 

In healthy subjects, voriconazole 200 mg twice daily for 30 days increased the maximum plasma levels and AUC of a single 60-mg dose of prednisolone hy 11% and 34%, respectively. ... 

No dosage adjustment of the corticosteroid is said to be necessary if voriconazole is given with prednisolone, and this also appears to be the case if voriconazole is given with hydrocortisone. ... 

In a recent study by Vogt et al. [67], 2D heteronuclear and homonuclear correlation SSNMR experiments involving H, C, F and P were used to demonstrate the inclusion of drug within the CD cavity in a variety of powder samples. The methods used in that work were similar to those applied in other studies of amorphous solid pharmaceutical dispersions, molecular cocrystals and molecular solid solutions [72,81]. In [67], the examples were shown for the drugs diflunisal, adefovir dipivoxil, voriconazole, dexamethasone and prednisolone in complexes with aCD, pCD and sulfobutylether-substituted pCD. One of the systems examined was a 1 2 molar mixture of diflunisal (Scheme 4.2) and pCD lyophilized from a phosphate-buffered solution with a pH of 7.0. In addition to SSNMR spectra, 2D and HETCOR were performed.
