Post-script

As a salutary post script to the discussion that has been presented above, a final example of innovation is presented, an example which has caused a profound change in direction in the search of anti-AIDS drugs. Until a little over a year ago, all research into inhibitors of reverse transcriptase (RT, see above) relied upon mimicry of the nucleoside substrates in the polymerase reaction. Here is an enzyme for which the reaction pathway is known only in bald outline and for which there is no structural information. Janssen s approach was masterly [84]. A library of 600 molecules, each prototypes of different chemical series and without activity in standard pharmacological assays, were screened for anti-HIV activity in vitro. It was discovered that (45) had modest but specific anti-HIV activity and lead optimisation eventually uncovered (46) and (47) as representatives of the TIBO series. It was subsequently shown that these compounds act as non-competitive inhibitors of RT, an indication that even if structural data were available, they would be useless in developing the series towards a drug candidate. As if by coincidence, several...

With the platform mentioned in 3.6, we have performed the post-synthesis verification procedure for several examples from the three scenarios mentioned. We here present results for one example for each scenario. EXAMPLEl is a working example performed by the AMICAL system. EXAMPLE2 is a part of the IKS pseudocode, where the register transfer description has been extracted by hand from microcode and a logic level implementation. EXAMPLES is a basic block from a differential equation solver performed by SUSAN. The proof script containing proofs of lemmas as well as HOL level and meta level decision procedures consist of about 2000 lines of SML code. 

Procedure The children were taken to a quiet area where the computer was set up. It was controlled by a mouse only, no keyboard being used. None of the children had encountered this type of computer before. The general scheme involved two sessions within one week of either individual or peer work, and an individual post-test for all, one week later. The children were first introduced to the problem via a practice task, lasting about ten minutes, in the course of which the main features of the problem and the interface were introduced. This phase was a tightly scripted tutorial situation very much under the control of the experimenter. Then the main task was presented, and the children were left to work on it with minimal experimenter support for a period of 30 minutes. On the second session, the children were rapidly presented with the introductory task and then had 25 minutes to work on the main task, unless they succeeded before this. On the individual post-test session, the same main task was used but with a different initial state (location of the characters and the transport) so that the subjects could not succeed by directly replicating their previous responses. Twenty minutes was the maximum time allowed on this session.
