Poly-L-lysine solution

Poly-L-lysine solution (Sigma Chemical Co., St. Louis, MO). 

Immerse the slides for 30 min in a large staining dish containing a 1 10 dilution of 0.1% poly-L-lysine solution in deionized water see Note 5). Remove the slides and oven-dry for 1 h at 60°C. 

Vacuum deposition and annealing of PSFs were performed as it was reported earlier [2]. Poly-L-lysine, cosine and hematoxylin were purchased from SIGMA-ALDRICH and used without purification. Poly-L-lysine (1 mg) was dissolved in 1 ml of distilled water (pH 5.0) and mixed with cosine aqueous solution (6x 10 m/1). Eosin-poly-L-lysine solution was deposited on PSF and on a bare... 

The poly-L-Lysine solution present in the wells is discarded and the wells are blocked with 200 p,L of TBST-Casein for 1 h at room temperature. 

Coat clean glass microscope slides with poly-L-lysine by pipeting 2 mL of an aqueous 10 pg/mL solution of poly-L-lysine (8-14 kDa) onto the slides. Incubate for 3 h and decant the poly-L-lysine solution.

Dip slides into a poly-L-lysine solution (slide adhesive solution, 0.1% w/v in H O Sigma P 8920). After withdrawal of the rack, the solution should wet the glass surface uniformly and stay on slides. Air-dry slides and store at 4°C. 

Drain well (poly-L-lysine solution can be reused). Rinse ten times in pure H O. 

To coat a plastic surface, the poly-L-lysine solution need only be 100 lg/ml. Since most plasticware cannot be baked or autoclaved after coating, use filter-sterilized poly-L-lysine.
