The 7-0-hemlsucclnate-7-desacetyl derivative of forskolin has been coupled to Sepharose to form an affinity resin. 2 Solubilized adenylate cyclase from rat brain and heart, but not pigeon erythrocytes, binds to this resin and can be eluted from the resin when forskolin is included in the buffer. Preparations of adenylate cyclase devoid of Ns activity are recovered from this column consistent with the proposal that forskolin binds to the catalytic subunit(s). Adenylate cyclase preactivated with guanosine-5, 3>Y i idodiphosphate (GppNHp) also binds to the forskolln-Sepharose column and elutes from the column as a complex of the catalytic subunlt(s) and the activated Ns protein.

Wheeler et have confirmed that for concentrations of Na above 20 mM, the Na -glycine symporter of pigeon erythrocytes is divalent with respect of Na", but below 20 mM Na, the system is monovalent with respect of Na". They also found that the Na -L-alanine symporter of pigeon erythrocytes and rabbit reticulocytes was monovalent with respect to Na over a wide range of Na concentration, and that the Na" "-linked symporter that translocates a-aminoisobutyric acid in Ehrlich ascites cells is similarly monovalent with respect to Na. ... 

One of the major operational differences between S and Ns is that in erythrocytes and certain other tissues a ubiquitous soluble protein (cytosolic factor or CF) is required to bind a nucleotide to the S site efficiently. Only about 10% of the Ns of pigeon erythrocyte membranes can be ADP-ribosylated in the absence of CF. We had known of this soluble protein for several years before Enomoto proved that it was concerned only with the proper loading of the S site with a guanyl nucleotide [1]. Membranes so loaded with GTP7S or Gpp(NH)p could be washed and subsequently ADP-ribosylated without any further supply of nucleotide or of CF.

The problems of identification are not nearly so acute when dealing with metalloproteins as they have a wide range of g values, and a number of these have been observed in whole tissue. Thus, the low spin ferric form of cytochrome P-450 has been observed in liver and its concentration found to be reduced in Morris hepatomas [82]. Adrenal ferredoxin in adrenal glands [83], sulphite oxidase in liver [84], iron-sulphur proteins in pigeon heart [85], methaemoglobin and erythrocyte catalase in whole blood are further examples in mammals.
