Peptide-loaded liposomes

Physically crosslinked HA hydrogel containing vasoactive intestinal peptide-loaded liposomes was developed for the treatment of endotoxin-induced uveitis. Interactions between HA chains and liposomes increased the viscosity of the gel and resulted in a delayed release of the vasoactive intestinal peptide [87]. However, the low stability of HA microparticles and physically crosslinked HA hydrogel is a limitation for protein delivery applications. [Pg.167]

McLaughin and coauthors have developed a straightforward method to assess the binding of proteins or peptides to liposomes (Buser and McLaughlin, 1998). They use sucrose-loaded hposomes obtained by extrusion through 0.4 fim pore size filter. When such hposomes are mixed with proteins in an aqueous buffer, they can be readily pelleted by ultracentrifugation. By comparing the amount of protein in the pellet and in the... [Pg.104]

To review, in an experimental mouse model, LPDI/E7 vaccination both prevents the establishment of metastatic E7-expressing tumors in naive mice through an induced E7-specific T-cell immune response and, in mice with previously established E7-expressing tumors, causes tumor regression with one subcutaneous injection of LPDI/E7 [Han SJ, et al. Subcutaneous antigen loading of dendritic cells by liposome-protamine-DNA (LPD) nanoparticles results in their activation and induction of specific antitumor immune response (impublished)]. A robust immune response follows administration of LPDI/ peptide particles, which can be used as either a preventative or therapeutic cancer vaccination strategy due to the ability of the particles to prevent and eliminate tumors, respectively, in mouse models. [Pg.250]

In designing polymer-coated liposomes containing peptide drugs, attention should also be paid to their structure. Drugs can be loaded into a liposomal... [Pg.175]
