PDA medium

Test for Yeasts and Molds The plate method is used for testing molds and yeast in semisolids. The procedure is similar to that of the total count test. Instead of SBCDA medium, Sabouraud dextrose agar (SDA) medium or potato dextrose agar (PDA) medium is used. Samples are incubated for five to seven days at 20-25 °C to identify the presence of yeasts and molds. [Pg.286]

Phenol Coefficient Test. The first important attempt at standardizing testing methods was known as the phenol coefficient test (96). It has been modified several times, and is an official AO AC screening test recognized by EPA and PDA. The phenol coefficient test compares the activity of disinfectants to that of phenol, under specific conditions, to give a number that measures the activity of the chemical tested with respect to that of phenol, ie, the phenol coefficient. Fresh inoculum can come from stock culture dishes kept under refrigeration. If these should become depleted, healthy strains of mycelium from the crop cultures can be used to inoculate sterilized agar media in dishes. To do so shake the crop culture jar violently to break up the mycelium. The inhibition of A. parasiticus growth by [4] was determined in our laboratory using a modification of the assay reported by Amoldi and coworkers (M) stilbene was dissolved in acetone and appropriate amounts were added to sterile potato dextrose agar (PDA) at 50 C to give concentrations of 10, 20, 50, and 100 ug/ml. The medium was dispersed in 60 mm tissue culture dishes and inoculated with 4 mm PDA plugs of actively growing A. parasiticus cultures. Plates were incubated in the dark at 30 C and colony diameters were measured daily. The reaction proceeds by the dissociative adsorption of acetic acid to form acetate ligands. Elthylene subsequently inserts into a Pd-acetate bond. The cation is then reduced by the reaction to form the neutral Pd°. The reaction is analogous to the Wacker reaction in which ethylene is oxidized over Pd + to form acetaldehyde. Pd° is subsequently reoxidized by oxygen to form pd2+[35,36,44]... [Pg.290]
