Hydroxyacetanilide. This derivative (21), also known as 4-acetamidophenol, acetaminophen, or paracetamol, forms large white monoclinic prisms from water. The compound is odorless and has a bitter taste. 4-Hydroxyacetani1 ide is insoluble in petroleum ether, pentane, and ben2ene slightly soluble in diethyl ether and cold water and soluble in hot water, alcohols, dimethylformamide, 1,2-dichloroethane, acetone, and ethyl acetate. In order to avoid citation of difficult chemical names, generic names came into being. However, in the beginning different names were independently assigned to the same substance in different countries. For example, not everybody would know that acetaminophen, 7V-(4-hydroxyphe-nyl)acetamide, 4 -hydroxyacetanilide, p-acetamidophenol, A -acetyl-p-aminophenol, acetomenophen and paracetamol are the same substance (see Fig. 41.2). 

Toward understanding the mechanistic action of toxicology using 2DE, Foun-toulakis et al. showed the hepatotoxic effects of paracetamol (acetaminophen) overdose in human and rodent liver [74]. They found that the expression of 35 proteins was altered after treatment with paracetamol or its nontoxic regioisomer 3-acetamidophenol. Paracetamol selectively increases or decreases the phosphorylation state of proteins. This effect translates to a decrease in protein phosphatase activity [80]. The control of cellular functions of cells may be lost as a result of the dephosphorylation of certain regulatory proteins due to paracetamol overdose.
