Therefore, we applied FL to a component vaccine and attempted to solve the above problems. FL can deliver the encapsulated soluble protein directly into the cytosol of cultured cells and introduce it into the class I MHC antigen-presentation pathway. Moreover, a single immunization with ovalbumin (OVA) encapsulated in FLs but not in simple liposomes results in the potent priming of OVA-specific CTLs. Thus, FLs function as an efficient tool for the delivery of CTL vaccines.

Fig. 2 (A) C57B1/6 mice were immunized subcutaneously on days 0, 21, and 35 with 10 pg OVA and 0.3 pg MPLA either in ICMVs or soluble form. Assessment of the frequency of OVA-specific T-ceUs among peripheral blood mononuclear cells indicated strong induction of antigen-specific CTLs in response to ICMV vaccination. The role of adjuvant effects of saponins on specific T cell populations was studied by depletion of T cells experiments of QS-21 in mice [31]. Different formulations of saponin-adjuvanted vaccines were tested for humoral and cell-mediated responses in mice. Many antigens, often including hen egg albumin (ovalbumin, OVA) as the antigen to be tested, have been used for immunizations, enzyme immunoassays, antigen-specific CTL and cellular proliferation assays [31].
