Orlistat® Xenical

Lipase Inhibitor Orlistat (Xenical, Roche) is prescribed for the treatment of obesity. It inhibits the gastrointestinal lipase enzymes by binding to the lipase through the serine site and inactivates the enzyme. Fat in the form of triglycerides cannot be hydrolyzed by the lipase and converted to free fatty acids and monoglycerides. Thus, there is no uptake of fat molecules into the cell tissue.

Other diet pills. Other diet pills are classified in various ways. Orlistat (Xenical) is not classified as a controlled substance. A prescription is required. 

The two best-known prescription diet pills being sold today are sibutramine (brand name Meridia) and orlistat (Xenical). Sibutramine is an appetite suppressant. Orlistat prevents the intestines from absorbing fat. Other medications exist, but they are less common, and people are only supposed to use them for short periods of time. 

As previously discussed, two weight loss medications are currently available by prescription. Sibutramine (Meridia), which suppresses appetite, is approved for use in obese patients age 16 or older. Orlistat (Xenical), a fat blocker, is approved for children 12 and older and adults. Candidates for these medications are not simply people who want to lose a few pounds, even if their doctors tell them they should lose weight. A patient must be severely overweight and meet certain other criteria before a doctor will consider prescribing medication. 

Two other irreversible inhibitors are penicillin G (4.18), a /3-lactam antibiotic, and orlistat (Xenical or Alii, 4.19), an antiobesity drug (Figure 4.19). Penicillin G inhibits cell wall synthesis in bacteria, while orlistat inhibits the breakdown of fats in the small intestine.16 Both drugs contain acid derivatives in a strained four-membered ring. Nucleophiles in the active sites of the inhibited enzymes attack the reactive carbonyl groups and open the strained ring in an energetically favorable, irreversible process. 

Orlistat (Xenical /Roche) is a reversible gastric and pancreatic lipase inhibitor. The compound has no effect on appetite suppression but rather acts by inhibiting dietary fat absorption from the GI tract. Sibutramine (Meridia /Abbott) and its major active metabolites are re-uptake... 

The drug orlistat (Xenical), which is promoted for the treatment of obesity, acts by inhibiting pancreatic lipase, thereby reducing the digestion and absorption of fat in the smaU intestine. 

Such as sibutramine, a.k.a. reductil, Ectiva, Reduxade, Meridia, and so on, and orlistat Xenical. 

Orlistat (Xenical) inhibits the action of the enzyme lipase in the intestine, thereby inhibiting the digestion and absorption of fats. This can lead to significant weight loss, but is often accompanied by flatulence, diarrhoea and oily stools. Its clinical application is described in Chapter 8.
