Muscarinic receptors alcohol

The alkaloid, cicutoxin, is distributed throughout the plant, though concentrated in the roots. Cicutoxin is an unsaturated aliphatic alcohol which is postulated to exert toxicity by central cholinergic stimulation. Specifically, stimulation of both nicotinic and muscarinic receptors occur. 

Binding to, and irreversible inhibition of, muscarinic receptors (selectively M3 compared with Mj) was observed with some iV-(chloro- or bromoethyl)piperidines, and shown by kinetic studies of their hydrolysis, to be related to the intermediate azoniaspiro[2,5]octanes (Scheme 17) <92MI 840-01, 93MI 840-02>. The aziridinium ion is the most potent compared with the parent piperidines or with the alcohol (143). 

As part of a medicinal chemistry program [16] directed toward discovering new muscarinic receptor ligands, a series of endo- and exo-2-alkyl-2-aza-bicyclo[2.2.1]heptan-5-ol (38 and 39) and endo- and exo-2-alkyl-2-aza-bicyclo[2.2.1]heptan-6-ol (40 and 41) esters have been synthesized from 2-alkyl-2-azabicyclo[2.2.1]heptan-5-enes (Scheme 2.4). Oxidation of the olefin and stereochemical manipulation of the secondary alcohols followed by esterification of the azanorbornene products provided the different isomers required for systematic investigation of the structure-activity relationships of this structural series.

Bucherer-Bergs products were also the starting points for the preparation of potent and selective muscarinic M3 receptor antagonists (Table 20) [83]. Hydantoins of general structure 228 (Scheme 40) participated in Mitsunobu reactions with alcohols to provide derivatives 229, which were reduced with sodium bis (2-methoxyethoxy)aluminum hydride (Red-Al) to afford imidazolidinones 230. Removal of the benzyl group allowed for further functionalization of the secondary amines to yield compounds 231.

S)-Hyoscyamine and (6)-scopolamine are esters of the amino alcohols tropine and scopine with ( S)-tropic acid, which is derived from phenylalanine (Fig. 3). The two alkaloids occur exclusively in the Solanaceae family. They act as antagonists of muscarinic acetylcholine receptors (parasympatholytics) and lead to an increase in pulse rate, relaxation of smooth muscles, e.g., in the gastrointestine and the bronchial tract, reduction of salivary, bronchial, gastric, and sweat gland secretion. While hyoscyamine is a central stimulant, scopolamine depresses the central nervous system.
