Probably, one of the most valuable advances in this field has dealt with the first chemoenzymatic synthesis of the stable isotope-enriched heparin from a uniformly double labelled 13C, 15N /V-acetylheparosan from E. coli K5. Heteronuclear, multidimensional nuclear magnetic resonance spectroscopy was employed to analyze the chemical composition and solution conformation of N-acety 1 hcparosan, the precursors, and heparin. Isotopic enrichment was found to provide well-resolved 13C spectra with the high sensitivity required for conformational studies of these biomolecules. Stable isotope-labelled heparin was indistinguishable from heparin derived from animal tissues and might be employed as a novel tool for studying the interaction of heparin with different receptors.30... [Pg.337]

Additionally, in part because of the pervasive intrusion (or timely arrival) of microprocessors, tools for structure determination are constantly being refined and enlarged in scope. Examples include multidimensional nuclear magnetic resonance (NMR), which is now commonly used for large molecules (the first commercial NMR spectrometers were introduced in the late 1950s) and, even more recently, near-field microscopy, which uses a lensless technique for VIS spectroscopy and thus sidesteps the normal problems of resolution by accumulating structural features a little at a time, is being developed (the first compound microscope became available in 1610). ... [Pg.47]

Multidimensional nuclear magnetic resonance (NMR) techniques, since their inception in the 1970s, have turned into a powerful tool that provides structures and dynamics of very complex molecules, crystals, and proteins. By extending these ideas to the optical regime, it is possible to come up with novel classes of spectroscopies that can probe complex vibrational motions. Its realization, however, has to wait for the recent breakthrough in femtosecond laser techniques, which has provided necessary impulsive excitation to overcome tremendous line broad-... [Pg.561]
