In our previous work [63], we studied the hydrolysis kinetics of lipase from Mucor javanicus in a modified Lewis cell (Fig. 4). Initial hydrolysis reaction rates (uri) were measured in the presence of lipase in the aqueous phase (borate buffer). Initial substrate (trilinolein) concentration (TLj) in the organic phase (octane) was between 0.05 and 8 mM. The presence of the interface with octane enhances hydrolysis [37]. Lineweaver-Burk plots of the kinetics curve (1/Uj.] = f( /TL)) gave straight lines, demonstrating that the hydrolysis reaction shows the expected kinetic behavior (Michaelis-Menten). Excess substrate results in reaction inhibition. Apparent parameters of the Michaelis equation were determined from the curve l/urj = f /TL) and substrate inhibition was determined from the curve 1/Uj.] =f(TL) ... [Pg.570]

The full capacity of the lipase-mediated technique for C-terminal deprotection is demonstrated in the synthesis of complex C>-glycopeptides, which are sensitive to both acids and bases, The key starting material, serine glycoside heptyl ester 11, is selectively depro-tected at the C-terminus by lipase obtained from the fungus Mucor javanicus (Scheme 22). The free carboxylic acid derivative 12 is coupled with an N-terminally protected glyco-dipeptide 13 to generate the tripeptide 14. [Pg.309]

In addition to these examples, the lipase from Mucor javanicus accepts various further glycosylated amino acids and peptides as substrates and selectively deprotects them at the C-terminus (Fig. 13). The enzyme tolerates varia-... [Pg.80]

Interesterification can also be catalysed by enzymes, many of which show useful specificities. The 1,3-specific lipases, such as those derived from Aspergillus niger, Mucor javanicus, M. miehei, Rhizopus arrhizus, R. delemar, and R. niveus, are particularly useful for interesterification. They are used to effect acyl exchange at the sn- and 3 positions while leaving acyl groups at the sn-2 position unchanged. [Pg.293]
