The chemical composition of caramel color is not yet fully understood but some compounds identified in the low weight fraction are considered caramel markers. All caramel classes contain 5-hydroxymethyl)-2-furaldehyde (5-HMF). In caramel classes in and TV, 4-methyUmidazole (4-MeI) has been detected, while 2-acetyl-4(5)-tetrahydroxybutylimidazole (THI) was found only in class HI caramel colors. The analysis of five caramel III samples by SPE/HPLC-MS revealed concentrations between 28.3 and 46.8 iglg THI and 73.3 to 187.8 for 4-MeP (see Figure 5.2.3). [Pg.337]

The researeh on dehydroepiandrosterone (DHEA) is limited beeause of the laek of radiolabeled metabolites. Robinzon et al. [126] showed that, using pig liver mierosomes, the radiolabeled metabolites of DHEA can be prepared in stable, pure form for bioehemical smdies. They utilized pig liver microsomal (PLM) fractions to prepare pH]-labeled 7a-hydroxy-DHEA (7a-OH-DHEA), 7[3-hydroxy-DHEA (7P-OH-DHEA), and 7-oxo-DHEA substrates from 50 pM [1,2,6,7-3H]DHEA. The metabolites were separated by silica gel PLC plates using ethyl aeetate-hexane-gla-eial aeetic acid (18 8 , v/v) as the mobile phase, extracted with ethyl aeetate, and dried under a stream of nitrogen. The purity of markers was determined with the use of TLC and GC/MS. [Pg.231]

A number of biochemical markers not associated with the cell envelope allow the specific detection of individual microorganisms in environmental samples. These include secondary alcohols. For example, Mycobacterium xenopi can be detected through the hydrolysis of wax ester mycolates, which liberates 2-docosanol, a characteristic and dominant secondary alcohol, which can be detected at low levels by GC-MS. This biomarker was found to be very useful for the rapid detection of M. xenopi in drinking water (159,160). Results from the GC-MS detection of 2-docosanol were obtained within 2 days compared to the 12 weeks required for culturable detection of M. xenopi. The detection limit for this type of approach was found to be 10 colony-forming units (CFU) ml" drinking water. [Pg.390]
