Poly(ethylenglycol)-a-distearoyl phosphatidylethanolamine,-w-maleimide FW 3,400 (mPEG-DSPE-maleimide, from Shearwater Polymers, Huntsville, AL). Dissolve mPEG-DSPE-maleimide in DMSO at 10 mg/mL immediately before reaction. 

To make scVEGF-lipid conjugate, add mPEG-DSPE-maleimide directly to deprotection reaction to a final molar protein-to-lipidratio of 1 2. Residual DTT will not affect modification reaction. Incubate lipidation reaction mixture at room temperature for 1 h.

Also, we attempted a localized transfection of tumor cells by the direct intratumoral administration of sterically shielded with pH-sensitive (containing mPEG-HZ-PE, 25) or pH-insensitive (containing mPEG-DSPE) conjugates TATp-liposome-pGFP complexes into the tumor tissue by the intratumoral injections. 

Weigh 2-4 mg of mPEG-DSPE-maleimide, and dissolve it in DMSO to make a final concentration of 10 mg/mL (2.9 mM). Add mPEG-DSPE-maleimide directly to Trout s reaction for a final protein-to-lipid molar ratio of 1 2. Incubate the reaction at room temperatnre for 1 h.

In addition, conventional PEG-linked liposomes (MPEG-DSPE liposomes) were prepared by adding MPEG-DSPE in the lipid mixture before hydration or after preparation of the bare liposome dispersion. In the latter method, the bare liposome dispersion was mixed with MPEG-DSPE and incubated at 50 °C for 3h to allow the anchor (i.e. distealoyl) moiety to be inserted into the liposome membrane. The final lipid concentration was maintained at 37.5 mM. 

In the case of MPEG-DSPE liposome, amount of MPEG-DSPE incorporated was determined as by Alien et al. Briefly, the liposome dispersion was ultracentrifuged (150000 G, 20 min) to obtain a supernatant containing MPEG-DSPE free of liposomes. Then, the MPEG-DSPE concentration in the supernatant was determined by a Bio Rad Protein assay method. From this value, the amount of liposome-incorporated MPEG-DSPE was calculated. 

Changes in the physicochemical properties of MPEG-SHz liposomes incubated in PBS at 37 C were evaluated with time, and compared with those of liposomes with stably linked PEG, namely, stabilised MPEG-SA liposomes and MPEG-DSPE liposomes. The lipid composition and linkage between PEG and the liposome surface of the liposomes tested here are shown in Table 1.

Egg phosphatidylcholine (EPC), cholesterol (Ch), mPEG ggg-DSPE, l,2-dioleoyl-3-trimethylammonium-propane (DOTAP), Rhodamine-PE (Rh-PE), and phosphatidylthio-ethanolamine (DPPE-SH) (all from Avanti Polar Lipids). 

A mixture of PC Chol (7 3), TATp-PECj j -PE, Rh-PE and either mPEG -HZ-PE (pH-sensitive) or mPEG gg-DSPE (pH-insensitive) at molar ratio 10 0.25 0.1 15 is evaporated under reduced pressure (see Note 3).
