Topiramate is a sulfamate-substituted monosaccharide derived from fructose with a broad spectrum of AED activity. It is approved for monotherapy or as an adjunct drug for partial or primary generalized tonic-clonic seizures in patients older than 10 years, as adjunct therapy in children aged from 2 to 10 years with partial-onset seizures, and in persons older than 2 years with Lennox-Gastaut syndrome (40,53). Topiramate also is approved for the prophylaxis of migraine headaches. [Pg.787]

Mishnick and co-workers have developed several techniques for determining the positions of substitutions on monosaccharide units and distribution of substituents along deriva-tized polysaccharide chains, primarily cellulose derivatives. Examples are found in references [61,62,63,64,65,66,67,68,69,70]. An important method involves perdeuteromethylation followed by partial acid-catalyzed hydrolysis, then reductive amination with propylamine, and finally permethylation to yield completely O- and A-alkylated charged oligosaccharides that are analyzed by MALDI-TOF-MS [68,70]. The derivatives used for GLC are indicated by methyl, acetyl, TFA (trifluoroacetyl) and TMS (trimethylsilyl) at the head of the list of compounds separated. All available hydroxyl groups are assumed to be substituted except in the case of partial or mixed derivatives where the positions of substitution are given under the appropriate monosaccharide. [Pg.249]
