In conclusion, the concept of solid-supported chemistry has been extended from the preparation of peptides and other biomolecules to any organic molecule. PS-and PEG-based resins are the most widely used however, syntheses can be carried out in any solid support. The development of new solid supports and linkers/ handles is crucial to fulfill the new requirements of modem drag discovery programs. 

The traditional method of drag development, at least in this century, has been to develop leads by first using, and then by isolating and identifying, the active chemical constituents from natural products, some of which may have been medicinally in use since antiquity. With the advent of modem organic chemistry some of these purified compounds were used directly (e.g., morphine, cocaine, atropine, quinine), and, once their chemical structures were ascertained, they became leads for hoped-for chemical modifications to achieve improved efficacy, less toxicity, or, at least, higher potency (e.g., dihydromorphinone, homatropine, acetylsalicylic acid).

Modem anti-inflammatory drags are very safe and inexpensive. Willow bark is difficult to take in a controlled manner, and does not inhibit blood clot formation, so it has no importance as a modem pharmaceutical. This plant was very important in developing modem drags, but its direct use wottld be a huge step backward. 

The first modem advanced drag delivery systems were developed by major pharmaceutical manufacturers such as Smith Kline French Laboratories, and with the growing recognition of the importance of advanced drag delivery technology there was a trend in these companies towards the establishment of special formulation units in their R D divisions. This sector soon attracted the attention of pharmaceutical entrepreneurs who saw opportunities for specialist formulation companies. By the late 1970s there were a number of such companies in operation, including Alza, Elan, Eurand and Pharmatec International. 

The early 1950s began the modem era of the treatment of anxieties and neuroses. We moved from the use of simple daytime sedation with dmgs that were essentially hypnotics, albeit at much lower subhypnotic doses, to compounds that became known as minor tranquilizers—the various polyol carbamates. In addition, the development of major tranquilizers that, for the first time in history, had more than just a custodial impact on schizophrenic-psychotic patients occurred simultaneously. These initial anxiolytics, with meprobamate now considered the prototype, were the first drags whose pharmacologic profile included more than just CNS depression.
