In 2007, Moda et al. [60] developed a set of Hologram QSAR models for a much smaller data set of 250 molecules in comparison to the above-discussed models. They found that using all atoms, bonds, connections, and chirality to define molecular fragments led to a set of encouraging models and the best one was achieved when the fragment size was from 4 to 7, which had q2 of 0.7 and r2 of 0.93. This model was too good to be true since the standard error was only 7.60, much smaller than the average experimental error, which is 14.5 based on 367 experimental data [55]. [Pg.115]

It is obvious that the available prediction models cannot give reliable estimations for oral bioavailability. The prediction errors were typically larger than 20% except the Moda s model, for which the data set only had 250 molecules in contrast, all other models utilized a much larger data set. [Pg.115]

Recently, hologram quantitative structure-activity relationship (HQSAR) was conducted by Moda et al. [63] on a series of structurally diverse molecules with known PPB. The best statistical model (n — 250, r2 = 0.91, and q2 — 0.72) was used to predict the PPB of 62 test set compounds, and the predicted values were in good agreement with the experimental results (ntest —62, q2 — 0.86, RMSEtest = 12%). It is indicated that this model used a much smaller data set than the VolSurf and Wang7s models. [Pg.116]

Generally speaking, PPB is an easier ADME-Tox property to be modeled in comparison to HIA and bioavailability. The prediction errors were only marginally larger than the experimental errors (Wang s model and Moda s model). When PPB is expressed as log Kt SA, the RMSEs were about 0.35 to 0.45. [Pg.117]
