Antibiotics The clinical use of three antibiotic cancer chemotherapeutic drugs (mitomycin C, mi-thramycin, and doxorubicin) has been associated with the development of nephrotoxicity. Mitomycin C is isolated from Streptomyces ca-espitosis and is used in the treatment of solid tumors. Renal failure (elevated BUN and serum creatinine concentration, proteinuria) induced by mitomycin C is dose dependent and cumulative. When administered alone, the incidence of nephrotoxicity is less than 1%, but in combination with 5-fluorouracil nephrotoxicity occurs more frequently and can be marked. The chemical species responsible for mitomycin C nephrotoxicity appears to result from the formation of alkylating metabolites. [Pg.1489]

Mitomycin C is a carcinogen (sarcomas and other cancers) in mice and rats after intraperitoneal, intravenous, or subcutaneous injections. It is teratogenic in mice (including skeletal defects). It is nephrotoxic. It is mutagenic in sister chromatid exchange assays. [Pg.1703]
